Bacterial permeability; total uptake of lysine by intact cells, protoplasts, and cell walls of Micrococcus lysodeikticus.

چکیده
A preceding study has shown that the pooled uptake and retention of lysine occurs equally or greater in protoplasts than in intact cells of Micrococcus lysodeikticus (Britt and Gerhardt, 1958). In the course of these analyses, it was noted that the intact cells apparently contained considerably more lysine than that found in the free amino acid pool. In the series of experiments to be reported here, we sought to relate more quantitatively the lysine entry into the internal pool with the total uptake by the cell, and also to determine where and how in the cell structure this larger, nonpooled balance of lysine uptake was localized. The latter aim was approached by comparing the total uptake and release of lysine by masses of intact cells, protoplasts, and isolated cell walls of the organism.

Binding of bacitracin to cells and protoplasts of Micrococcus lysodeikticus.
Bacitracin was tritiated by the Wilzbach technique and purified by carboxymethylcellulose chromatography. Binding studies between [3H]bacitracin and Micrococcus lysodeikticus cells and protoplasts indicated that the cells or protoplasts were saturated at 2 X lo5 molecules per cell. This value correlates well with the number of CS5-isoprenyl pyrophosphate molecules found in the membrane and adds...
متن کاملLiberation and osmotic properties of the protoplasts of Micrococcus lysodeikticus and Sarcina lutea.
Stable protoplasts may be released from MiCrococcua Zysodeikticus and Sarcina Zutea by digestion of the cell-wall with lysozyme in sucrose or NaCl solutions having an osmotic pressure of some 25 atmospheres, but not in glycerol solutions of the same osmotic pressure. The stability of the protoplasts depends not only upon the depression of the water activity by the solute but upon an osmotic pre...
متن کاملThe isolation of protoplasts from Bacillus megaterium by controlled treatment with lysozyme.
Certain bacteria are killed and rapidly lysed following exposure to the enzyme lysozyme. Lysozyme sensitivity resulting in extensive lysis occurs in a relatively restricted number of species: notably Micrococcus lysodeikticus, Sarcina lutea, and certain Bacillus spp. It was shown some years ago (Epstein and Chain, 1940; Meyer and Hahnel, 1946) that a mucopolysaccharide extractable from the cell...
متن کاملDNA binding and uptake by nuclei isolated from plant protoplasts: factors affecting DNA binding and uptake.
DNA binding and uptake by nuclei isolated from soybean (Glycine max L. Merr.) protoplasts were investigated using radioactive homogeneous DNA prepared from soybean cells. DNA binding to nuclei was found to decrease drastically with increased incubation time. Total uptake and acid-precipitable uptake reached a maximum after 20 minutes of incubation. Optimum DNA binding and uptake occurred at pH ...
متن کاملImmunological properties of Micrococcus lysodeikticus membranes.
Membranes of Micrococcus lysodeikticus possess antigens which are distinct from other cellular components such as cytoplasm, ribosomes, and cell walls. Only a few (two to three) components are found when dissociated membranes are examined by immunodiffusion and immunoelectrophoresis techniques. Membranes treated with 0.3% sodium dodecyl sulfate, 0.3% Triton X-100, trypsin, phospholipase A or C,...
